Concatenated 16S rRNA sequence analysis improves bacterial taxonomy

Background: Microscopic, biochemical, molecular, and computer-based approaches are extensively used to identify classify bacterial populations. Advances in DNA sequencing bioinformatics workflows have facilitated sophisticated genome-based methods for microbial taxonomy although of the 16S rRNA gene is widely employed community as a cost-effective single-gene approach. However, sequence-based species identification accuracy limited by multiple copies their higher sequence identity between closely related species. The availability large volume whole-genome data provided an opportunity develop comprehensive species-specific reference libraries. Methods: were retrieved from whole genomes complete stage at Genome database. With defined rules, four copy variants concatenated library. similarity search was performed with web-based BLAST program, MEGA software construct phylogenetic tree. Results: Using this approach, libraries developed Streptococcus (S. gordonii, S. mitis, oralis, pneumoniae). Sequence analysis using yielded better resolution than single approaches. Conclusions: approach very effective classifying genetically may reduce misclassification genome assemblies.
